Sperm Analysis and Sperm Preparation

Sperm analysis and sperm preparation are important procedures conducted in the field of reproductive medicine to assess and optimize sperm quality for fertility treatments.

  1. Sperm analysis: Also known as semen analysis, is a laboratory test that evaluates various parameters of sperm quality. It provides valuable information about sperm count, motility (movement), morphology (shape), and other factors that can affectfertility.

During a sperm analysis, a sample of semen is collected through masturbation into a sterile container. The sample is then examined under a microscope by an Embryologist, Andrologist or a trained technician. They will assess the characteristics of the sperm, including concentration (number of sperm per milliliter of semen), motility (ability to swim forward), and morphology (shape and structure).

Sperm analysis helps provide insights into the potential causes of fertility issues and assists in determining the most suitable treatment options, such as intrauterine insemination (IUI) or in vitro fertilization (IVF).

  1. Sperm preparation: Sperm preparation is the process of isolating and preparing high-quality sperm for use in assisted reproductive techniques like IUI or IVF.

The procedure involves the following steps:

– Semen collection: Similar to sperm analysis, a semen sample is collected by masturbation into a sterile container.

– Semen liquefaction: After collection, the semen goes through a natural process called liquefaction, wherein it transforms from a gel-like state to a liquid form. This usually takes around 20-30 minutes at room temperature.

– Sperm washing: It is performed to separate the motile, healthy sperm from other components in the semen, such as seminal fluid and debris. It usually involves layering the semen sample onto a density gradient medium and centrifugation, which helps isolate the best-quality sperm.

– Sperm concentration and dilution: The isolated sperm are then concentrated to increase their numbers and diluted in an appropriate medium. This step helps ensure the optimal sperm concentration for the specific fertility treatment being performed.

– Sperm selection: This technique can include using the swim-up method, where the highly motile sperm swim upwards, allowing for their isolation and subsequent use.
